Chemical Identity
Orexin-B analytical reference compound. Molecular weight 2937 Da.
Research Overview
Orexin-B (Hypocretin-2) is a 28-amino acid neuropeptide derived from the same prepro-orexin precursor as Orexin-A. Produced by hypothalamic neurons, Orexin-B signals preferentially through the OX2R receptor, showing approximately equal affinity for OX2R compared to Orexin-A but markedly lower affinity for OX1R. This receptor selectivity profile makes Orexin-B a valuable tool for distinguishing OX2R-mediated effects in research settings.
Orexin-B lacks the disulfide bonds and pyroglutamyl modification that stabilize Orexin-A, resulting in a shorter biological half-life and more rapid degradation by tissue peptidases. Despite this, Orexin-B plays complementary roles in arousal regulation, metabolic control, and autonomic function. The OX2R receptor through which Orexin-B preferentially signals is particularly concentrated in the tuberomammillary nucleus (histaminergic wake-promoting neurons), making Orexin-B especially relevant to research on arousal and sleep-wake transitions. Preclinical studies have examined Orexin-B in the context of sleep architecture, hypothalamic-pituitary axis regulation, and metabolic signaling.

Research Applications
Orexin-B has been documented in the published scientific literature across the following research areas:
- OX2R receptor selectivity and binding studies
- Sleep architecture and arousal research
- Comparative orexin signaling investigation
- Hypothalamic-pituitary axis modulation studies
- Metabolic and feeding behavior research

Researchers should consult the primary literature for experimental protocols, concentrations, and conditions relevant to their specific area of investigation involving Orexin-B.
Storage and Handling
Orexin-B is supplied as a lyophilized powder and should be stored at -20°C upon receipt for long-term stability. Protect from light, moisture, and repeated temperature fluctuations. Allow the sealed vial to equilibrate to room temperature before opening to prevent condensation.
For reconstitution, add sterile water or an appropriate buffer slowly along the vial wall. Gently swirl to dissolve. Reconstituted Orexin-B solutions should be stored at 2-8°C and used within 30 days. Aliquoting is recommended. Consult the Safety Data Sheet (SDS) for detailed handling guidance.
For laboratory research use only (in vitro). Not for human or animal use. Not for diagnostic, therapeutic, or clinical purposes. Orexin-B is supplied as an analytical reference compound for use by qualified research personnel at accredited institutions. Prescott Bio Canada does not provide guidance on administration, dosing, or use in living organisms.
