GHK-Cu: Research Summary

Mechanism
GHK-Cu is a copper-binding tripeptide (Glycyl-L-Histidyl-L-Lysine complexed with divalent copper) originally isolated from human plasma. It functions as a copper-transport chaperone with high affinity for Cu(II), and copper delivery into cells appears to be central to many of its downstream effects. Reported activities include stimulation of collagen and glycosaminoglycan synthesis in dermal fibroblasts, upregulation of decorin and other matrix- remodeling proteins, activation of superoxide dismutase and other antioxidant systems, and broad transcriptional modulation. A widely cited Connectivity Map analysis identified GHK as a compound that reverses the gene-expression signature of aged skin fibroblasts toward a younger profile.
Published trials
The strongest body of clinical evidence is topical (creams, serums) for skin remodeling. Randomized split-face and vehicle-controlled trials of GHK-Cu- containing topicals report improvements in fine lines, skin firmness, thickness, and photodamage markers over 12-week horizons. Pre-clinical work covers wound healing (excisional and diabetic-wound rodent models), hair follicle stimulation (dermal papilla cell culture and follicle organ culture), and neuroprotection (rodent stroke and neurodegeneration models).
Systemic (subcutaneous or intravenous) GHK-Cu human clinical trial data is very limited. Copper-peptide formulations have been used in older wound- healing trials, but modern RCT-grade systemic-administration data is not available at the scale of the topical literature.
Safety profile
Topical GHK-Cu formulations have a favorable published tolerability profile, with occasional local irritation reported. Systemic safety data is much thinner. Copper toxicity is the theoretical concern with sustained systemic exposure — Wilson's disease and hepatic copper overload are the reference pathologies — but published pre-clinical work at typical research doses has not reported acute copper-loading events. GHK-Cu is not approved for human therapeutic use as a systemic drug by any major regulator; its regulated appearances are as a cosmetic ingredient.
How it compares in its class
GHK-Cu is the most-studied member of a small class of "copper-carrier peptides" used in cosmetic and pre-clinical regenerative research. Within regenerative peptides more broadly, its niche is skin remodeling and matrix- synthesis stimulation rather than the gastrointestinal-healing focus of BPC-157 or the actin-binding wound-closure profile of TB-500. It has the deepest topical clinical literature of any peptide in the class.
